FOX04-DRI is a clever piece of peptide engineering. It's modeled on a section of the FOXO4 transcription factor that normally talks to p53 inside aging ("senescent") cells. By building it with mirror-image amino acids in reverse order, researchers create a molecule that's much more stable in lab conditions than a natural peptide.
How it's been studied: Published work — notably from the Demaria lab — used FOX04-DRI as a research tool to break the FOXO4-p53 connection inside senescent cells, which then triggers them to clear out (a "senolytic" mechanism). All work has been in cultured cells and aged mice.
